KARAKTERISASI STRUKTUR SENYAWA KUMARIN GLIKOSIDA DARI BIJI BUAH RAMBUTAN (Nephelium lappaceum L.)
Abstract
Rambutan (Nephelium lappaceum L.) merupakan tanaman musiman yang banyak hidup di daerah tropis. Senyawa fenolik kumarin glikosida telah isolasi dari biji buah N. lappaceum. Penelitian ini bertujuan untuk mengetahui struktur senyawa kumarin glikosida pada biji buah N. lappaceum. Proses isolasi senyawa dilakukan dengan beberapa metode ekstraksi dan kromatografi. Fraksi yang sebagai sumber senyawa kumarin glikosida adalah fraksi etil asetat. Isolat murni yang diperoleh sebanyak 17,6 mg berbentuk kristal berwarna kuning dengan hasil uji fitokimia positif golongan fenolik. Data NMR-1H (CD3OD, 500 MHz) menunjukkan geseran kimia δ (ppm) 8,15 (1H, brs, H-2), 6,91 (1H, d, J=8,2 Hz, H-3), 6,77 (1H, s, H-6), dan 6,44 (1H, s, H-8), 5,57 (1H, s, H-1"™), 4,04 (1H, s, H-2"™), 3,85 (1H, dd, J=9,25;3,45 Hz, H-3"™), 3,49 (1H, t, J=9,55 Hz, H-4"™), 3,61 (1H, m, H-5"™), 1,27 (3H, d, J=6,15 Hz, H-6"™). Data TOF-ESI-MS m/z memberikan massa molekul 348,28 [M+Na+H]+. Berdasarkan hasil analisis senyawa yang diperoleh dari fraksi etil asetat biji buah N. lappaceum adalah senyawa 5-O-α-L-ramnosa-7-hidroksikumarin.
