ISOLASI DAN KARAKTERISASI SENYAWA FLAVONOID FRAKSI ETIL ASETAT KULIT BATANG DURIAN KLAWING (Durio graveolens Becc.)
Abstract
Senyawa flavonoid telah diisolasi dari fraksi etil asetat kulit batang durian klawing (D. graveolens Becc.). Tahapan isolasi senyawa flavonoid adalah ekstraksi, fraksinasi, kromatografi vakum cair (KVC), dan kromatografi kolom gravitasi (KKG). Struktur senyawa ditentukan menggunakan proton nuclear magnetic resonance (1H-NMR) (500 MHz), carbon nuclear magnetic resonance (13C-NMR) (125 MHz), heteronuclear multiple bond correlation (HMBC) dan electro spray ionization-mass spectrometers (ESI-MS). Pergeseran kimia senyawa flavonoid (δH ppm) : 3,92 (3H, s, OCH3); 6,34 (1H, d, J = 2,3 Hz, H-6); 6,54 (1H, d, J = 2,3 Hz, H-8); 7,14 (1H, d, J = 2,1 Hz, H-2"™); 6,87 (IH, d, J = 8,4 Hz, H-5"™); 6,94 (IH, d, J = 8,2 Hz dan 2,1 Hz, H-6"™); 8,12 (2H, 3"™-OH dan 4"™OH); 8,19 (IH, s, 3-OH); 13,02 (IH, s, 5-OH), pergeseran kimia (δC ppm) : 154,5 (C-2); 124,2 (C-3); 181,7 (C-4); 163,5 (C-5); 98,7 (C-6); 166,6 (C-7); 92,9 (C-8); 158,8 (C-9); 106,7 (C-10); 123,4 (C-1"™); 115 (C-2"™); 145,6 (C-3"™); 56,4 (OCH3); 146,3 (C-4"™); 117,1 (C-5"™); 121,4 (C-6"™), data ESITOFMS memiliki m/z 301,0736 [M + H]+ dan m/z 299,0583 [M - H]-. Berdasarkan kemiripan data spektrum dengan literatur, diketahui bahwa senyawa flavonoid tersebut adalah senyawa luteolin-7-metil eter atau 3"™,4"™,5-trihidroksi-7-metoksiflavon.
