KARAKTERISASI SENYAWA SITOTOKSIK DARI FRAKSI ETIL ASETAT DAUN KRATOM (Mitragyna speciosa Korth.) DAN AKTIVITASNYA TERHADAP SEL KANKER PAYUDARA T47D
Abstract
Senyawa sitotoksik sebanyak 3,1 mg berwarna hijau kecoklatan dan titik leleh 130-135 oC telah diisolasi dari fraksi etil asetat daun kratom (Mitragyna speciosa Korth.). Analisis sitotosik terhadap sel kanker payudara T47D menunjukan senyawa tersebut tergolong dalam sitotoksik moderat dengan nilai IC50 161,67 µg/mL. Karakterisasi senyawa sitotoksik menggunakan spektrofotometri Uv-Vis memberikan serapan maksimum pada panjang gelombang 284, 308, dan 318 nm yang menunjukan adanya transisi elektronik π→π*, n→σ*, dan n→π*. Data Uv-Vis menunjukan adanya kromofor C=O dan C=C. Analisis IR menunjukan serapan "“OH (3226-3029 cm-1), C=C aromatik (1557-1474 cm-1), C=O (1738 cm-1), C-O (1233 dan 1043 cm-1). Analisis 1H-NMR (CDCl3, 500 MHz) menunjukan δH 3,30-3,80 ppm (glukosa), δH 4,91 ppm (1H, d, J= 10,5 Hz, H-1"™"™), δH 6,88 ppm (1H, d, J=8,0 Hz, H-5"™), δH (1H, d, J=9,0 Hz, H-6"™) dan δH 7,68 ppm (1H, br). Berdasarkan hasil analisis Uv-Vis, IR, 1H-NMR serta uji metabolit sekunder diprediksi senyawa sitotoksik merupakan kuersetin-3-O-β-glukopiranosida.
