ANTIBACTERIAL ACTIVITY OF N-HEXANE EXTRACTED OF Ploiarium alternifolium LEAF

Abstract

This study was conducted to find out: chemical characteristics and antibacterial capability of the compounds extracted with n-hexane in leaves of Ploiarium alternifolium. The compound was obtained by extraction of n-hexane from P. alternifolium leaf and purified by vacuum liquid chromatography (VLC) and gravitational column chromatography (GCC) and which each characterized by thin layer chromatography (TLC). The antibacterial properties were determined from KCV fractions against Gram-Positive Staphylococcus aureus and Gram-Negative Shigella flexneri bacteria. Chemical characterization was performed by infrared spectroscopy. The result of this research is 7 fraction of KCV, that is A, B, C, D, E, F, and G. Furthermore, fraction D is purified by GCC. The pure compounds obtained were characterized by infrared spectroscopy. The pure compound has ester, methyl, methylene, methyl, cyclohexane and open-chain aliphatic groups. All VLC yield fractions exhibit antibacterial properties against Shigella flexneri, but do not inhibit the growth of Staphylococcus aureus bacteria. Inhibition of Shigella flexneri bacteria growth was highest by G (eluen ethyl acetate) fraction with a clear zone diameter ratio of 106% compared to clindamycin clear zone diameter (positive control). Fraction D has a clear-zone diameter ratio of 79% to 87% compared to the clindamycin clear zone diameter.
