Abstract

The ability of nipah worms to digest consumed feed depends on the presence or absence of appropriate enzymes and the composition of microbes present in the environment and digestive tract that can assist in the cellulose breakdown process. Cellulolytic bacteria can be found in weathered plant organic matter, such as wood and leaves, in mangrove forests. This study was conducted to characterize cellulolytic bacteria on nypa palm worm substrate based on colony shape and Gram staining. Bacteria were isolated using the Pour Plate Method with CMC selective media and suspensions were prepared using 0.9% physiological NaCl. The colony shapes obtained from cellulolytic activity screening showed that 13% had an irregular shape, 4% had a filamentous shape, and 83% had a circular shape. Gram staining results revealed that 96% of isolates were Gram-negative bacteria and 4% were Gram-positive. Among the isolates, 39% had a coccobacilli cell shape, 35% had a coccus shape, and 26% had a bacilli shape.
